This data is part of a multi-investigator, multi-disciplinary collaboration called the SCTLD resistance research consortium (RRC) investigating resistance and susceptibility among Orbicella faveolata populations off Broward and the Lower Florida Keys. These digitized histological slides include tissue biopsies from wild O. faveolata colonies from two regions (the Kristin Jacobs Coral Reef Ecosystem Conservation Area in Broward County, and Looe Key and Sand Key in the Florida Keys National Marine Sanctuary) and three resistance categories (“Low Resistance”, “Medium Resistance”, “High Resistance”) collected in late-May – mid-June, 2021. All slides were stained with hematoxylin and eosin and scanned on a digital slide scanner to a maximum magnification of 40x. All images are in SVS format. This dataset only includes the raw histological slide collection and associated metadata from biopsied coral colonies. Results from the analysis of the histological slides will be available from partner labs in separate associated datasets.

Purpose Stony coral tissue loss disease (SCTLD) is a lethal disease affecting Florida’s coral reefs since 2014 whose etiology and etiological agent(s) are still unknown at the time of writing. The purpose of this project is to identify functional characteristics of the SCTLD phenotype at a cellular level to gain insights into the pathological mechanisms governing this disease. Understanding the functional changes associated with disease progression and resistance will provide a foundation for developing new treatments and diagnostics for intervention and management of SCTLD and other coral diseases. These digitized slides were collected as part of the histological analysis performed by the Resistance Research Consortium (RRC), a multi-institutional and multi-disciplinary collaboration of scientists working to understand the mechanisms of susceptibility and resistance to SCTLD.

Histological sections were prepared by the US Geological Survey (USGS) under the direction of Aine Hawthorne using standard processing techniques including dehydrating tissues with a graduated series of alcohols, infiltrating tissues with paraffin wax, embedding tissues in paraffin blocks, sectioning tissues and mounting onto glass slides for staining. Staining with hematoxylin and eosin was performed by IDEXX BioAnalytics (Columbia, MO). Stained slides were scanned on a digital slide scanner at a magnification of 40x using EasyScanInfinity software.
