SPMC Water Quality Sampling & Weather Record (2 per week)

Bring from lab - Temperature probe, 125 mL poly bottle, 250 mL beaker.
Pull out pumphouse hose to ditch, turn on water - valve is low in SW corner of pumphouse.

Temperature
(Cole-Parmer Digital pH/TEMP/mV/ORP Meter with automatic temp. comp. 5938-10, SN C95003264), corrected to NIST-tracable HB Inst. Co. calibrated thermometer (calib. Date 6/98) SN: H-B-A 06994.
Fill beaker, put in temp probe, note temperature, add 0.7 C correction.
Rinse poly bottle 3X, fill.
Shut off water.

 Dissolved Oxygen
(YSI Model 55/12FT, SN:97K0196 AL)
Calibration of YSI 55 meter
Make sure sponge in calibration chamber is wet.
Insert probe into calibration chamber.
Turn instrument on by pressing ON/OFF button.
Let stabilize for 15 min.
Enter calibration menu by pressing both arrow keys at once.
Enter local altitude (0), press ENTER.
Calibration value (100% sat.) is displayed, with current DO reading.
When DO reading is stable, press ENTER.
Enter salinity in (30).
Press ENTER, instrument will return to normal operation.
Set probe into beaker with stir bar spinning.
Read DO in PPM.

PH
(Cole-Parmer Digital pH/TEMP/mV/ORP Meter with automatic temp. comp. 5938-10, SN C95003264)
Put pH probe and temp. probe in 7.0 buffer, adjust STD.
Probes in 10.0 buffer, adjust Slope.
Probes and stir bar in beaker, measure pH.

Salinity
(ENDECO Type 102 Refractometer, range 0-45 , SN 1020017)
Measure by putting water on the refractometer, point at light & read.
Note temp on side and correct for temp with table.

Chlorophyll
(Turner Fluorometer Model 112-000, SN E002220TO)
Put 25 mm GFF glass fiber filter into filter holder & attach to 60 mL syringe.
Fill syringe twice to 50 mL line & gently filter, rinsing and filling vials.
Place filter in centrifuge tube, add 10 mL 90% acetone (in hood).
Label with date & place in freezer.
Measure chla weekly using Turner fluorometer:
Shake tubes vigorously.
Centrifuge 5 min at full speed, transfer to cuvette.
Blank fluorometer with 90% acetone.
Read sample, add two drops HCl, read again.
Enter results in Chla notebook.

Vials: Fill 1 - 4 mL autoanalyser vial for NO3 analysis, 1 - 20 mL acid-washed polyethylene bottle each for NH4, PO4 analyses.
Label with date & freeze.

Meteorlogical Measurements
Read rain gauge and empty it, if necessary.
Read max & min temps, reset with magnet.
Estimate cloud cover (0-4), wind direction & speed.
Record data in sampling form.
